      目的:探讨8周有氧运动对APP/PS1转基因小鼠皮层AMPA受体亚型GluR1的作用,为运动促进脑功能提供实验依据。方法:2月龄清洁级APP/PS1双转基因小鼠和C57BL/6J小鼠各40只,随机分为APP PS1安静组(AS)、APP PS1运动组(AE)、C57安静组(CS),C57运动组(CE)。运动组小鼠进行8w的跑台运动,5d/w,60min/d。前10min运动速度15m/min,后50min运动速度18m/min,跑台坡度为0%。跳台回避实验检测其行为学变化,记录小鼠潜伏期、错误反应次数及达标率。免疫组化方法检测皮层突触后致密物PSD-95。Western blot检测皮层AMPA受体GluR1亚基单位的表达及GluR1-Ser831和GluR1-Ser845磷酸化位点的活性。结果:1)行为学:4月龄AS组与CE组行为学变化不明显;8周有氧运动使CE组和AE组的潜伏期延长、达标率增加(P<0.05)。2)4月龄APP PS1小鼠皮层PSD-95较正常对照组减少;8周有氧运动明显增加其皮层PSD-95含量。3)4月龄APP PS1小鼠皮层AMPA GluR1受体含量及活性较正常对照组变化不大。8周运动后CE和AE组其皮层受体表达及磷酸化水平均较安静组增加。结论:4月龄的APP PS1双转基因小鼠学习记忆整体行为学虽然无明显下降,但细胞水平皮层突触密度减少,PSD-95 的组分AMPA受体GluR1活性下降。8周有氧运动提高小鼠跳台回避能力,同时明显增加皮层PSD-95含量及GluR1含量和活性。

      Objective: To investigate the effect of 8 weeks aerobic exercise on the AMPA-type glutamate receptor subunit GluR1 in the APP/PS1 transgenic mouse cortex. Methods: APP/PS1 transgenic mice (2 months old) and C57BL/6J mice(2 months old) were randomly divided into 8 weeks exercise and sedentary respectively, namely APP/PS1 sedentary group (AS), APP/PS1 exercise(AE), C57 sedentary (CS) and C57 exercise(CE). The exercised mice were subjected to a treadmill exercise protocol at the speed of 15m/min on an 0% slope for 10min/d then 18m/min for 50min/d. after exercise, the behavioral change was detected by step-down fear conditioning. The brains were subjucted to immunohistochemistry and immunoblotting to analyze changes of postysynaptic densities-95 (PSD-95) and CluR1 respectively. Results:1)Behaviorally, although there was no significant changes in 4 month-old AS compared with CS, the exercised mice performed significantly better than their respective control cohorts in step-down fear conditioning(P<0.05). 2)The PSD-95 was decreased in cortex in 4 months old AS compared with those in CS. The AE and CE group exhibited a significant increase of PSD-95 compared with their respective control cohorts. 3) Although the protein levels of GluR1 in the cortex had no differences between AS and CS, the activity of GluR1 was decreased in AS. The expression of GluR1 and the phosphorylation level were significantly increased in AE and CE after 8ws aerobic exercise. Conclusion: There was no significant decrease in learning and memory ability in four months-old APP/PS1 transgenic mice, while PSD-95 and GluR1 activity were decreased at the cellular level. The protocol used was able to promote PSD-95 and GluR1 activity with likely implications in the exercise-induced mental health.
