      摘要:目的:探讨全身振动训练对骨质疏松后骨骼肌的改善及肌肉生长抑制素(MSTN)的影响,为更好的缓解骨质疏松提供运动医学的科学依据。方法:3 月龄雌性 SD 大鼠随机分为假手术组(Sham 组),去卵巢组(OVX 组),全身振动训练组(OVX-V组),每组10只。8 周去卵巢建模成功后,OVX-V组开始振动训练(7 d/周,2 次/d,15 min/次、间隔 10 min,振动频率50 Hz、振幅 0.9 mm),其余2组正常饲养。8 周 后,测定子宫湿重、子宫内膜厚度;ELISA检测E2、P、T4的变化;RT-qPCR和western blot检测MSTN 、Act RIIB、Akt、m TOR、FoxO1的表达。结果:与OVX 组相比,全身振动训练后大鼠体重减轻、抓力增加;子宫湿重、子宫内膜厚度增加,但无统计学意义;激素水平E2和T4明显升高;并且振动训练显著下调Akt, mTOR,p-Akt 和p-mTOR的表达,显著上调MSTN,ActRIIB, FoxO1 和它的磷酸化表达。结论:8 w全身振动训练对骨质疏松大鼠骨骼肌质量有改善作用,其机制可能是通过促进蛋白合成抑制蛋白降解。MSTN有望作为一种新的肌肉力量评价指标,联合振动训练应用于运动训练和运动康复领域中。

      Abstract: Objective: This study explored the effect of whole body vibration training on ovariectomized rats and myostatin (MSTN), and aimed to provide a better scientific basis for the better relief of osteoporosis. Methods: Three-month old female rats were randomly divided into sham group (sham group), ovariectomized group (OVX group) and whole body vibration training group (OVX-V, group) (n = 10). After 8-week successful ovariectomized modeling, OVX-V rats started vibration training (7 d/week, 2 times/d, 15 min/ time, 10 min interval, vibration frequency 50 Hz, amplitude 0.9 mm), the other two groups were kept normal. After 8 weeks, the uterine wet weight and endometrium thickness were tested; the changes of E2, P and T4 were measured by ELISA; the expressions of MSTN, Act RIIB, Akt, m TOR and FoxO1 were detected by RT-qPCR and Western blotting. Results: Compared with OVX group, the rat’s body weight and grip strength were increased after the vibration training; the wet weight and endometrial thickness of uterus increased but with no significance; the levels of E2 and T4 increased significantly; vibration training significantly downregulated the expressions of Akt, mTOR, p-Akt and P-mTOR and significantly upregulated the expressions of MSTN, ActRIIB, FoxO1 and their phosphorylation levels. Conclusion: Eight weeks whole body vibration training can improve the quality of skeletal muscle in osteoporotic rats. The mechanism may be by promoting the degradation of protein synthesis inhibitory protein. MSTN is expected to be a new index of muscle strength evaluation, and combined vibration training can be used in sports training and sports rehabilitation.
