      摘要:目的:探讨葡萄糖转运蛋白1(Glucose Transporter 1, GLUT1)基因rs841853 G/T多态位点作为评估和筛选长跑运动能力遗传标记的可行性。方法:于2009-2010年选取78名中国女子专业长跑运动员,采用基质辅助激光解吸附电离飞行时间质谱检测技术解析GLUT1基因rs841853位点基因型,采用递增负荷跑台运动试验获取运动员的生理机能指标,并检索追溯运动员截止到2017年各主项的个人最好成绩。结果:中国女子专业长跑运动员rs841853 G/T多态位点的基因型GG、GT和TT的频率分布为65.4%、29.5%和5.1%,等位基因G和T的频率分布为80.1%和19.9%,符合哈温平衡定律。基因型频率和等位基因频率与北京汉族人群存在显著差异(P<0.05,P<0.05)。GT+TT基因型携带者的VO2AT/VO2max、VE和5000米项目个人最好成绩显著高于GG基因型携带者(P<0.05,P<0.05,P<0.05)。结论:GLUT1基因rs841853位点的T等位基因可能是评估优秀长跑运动能力的基因标记,有待于通过扩大样本量和功能学研究给予进一步验证。

      Abstract: Objective: This study aimed at exploring the feasibility of glucose transporter 1 (GLUT1) gene rs841853 polymorphism as a marker for evaluating and screening athletic ability of long distance runners. Methods: Seventy-eight Chinese female long distance runners participated in this research during 2009-2010; the genotypes of GLUT1 rs841853 were analyzed by matrix-assisted laser desorption ionization-time of flight mass spectrometry, and physiology indicators were determined by graded exercise testing on treadmill; personal best until 2017 in main events were retrieved. Results: 1) The genotype frequencies of rs841853 of Chinese female long distance runners were 65.4%, 29.5% and 5.1% for GG, GT and TT, respectively, and allele frequencies were 80.1% and 19.9% for G and T, respectively, which distributions were in Hardy-Weinberg equilibrium. These genotype frequencies and allele frequencies were significantly different from those of Beijing Han population (HapMap-HCB) (P < 0.05). GT and TT genotypes carriers showed greater VO2AT/VO2max and VE, better personal best in 5000 meter running than GG genotype carriers (P < 0.05). Conclusion: The T allele of GLUT1 rs841853 would be a feasible genetic marker to evaluate and identify excellent long distance running performance, which needs to be further verified by further research with large sample size and function.
