      摘要:目的:探讨耐力运动和/或双氢睾酮(DHT)对大鼠肝组织AMPK和ACCβ表达的影响及机制。方法:24只雄性SD大鼠随机分为4组:运动组(E,n=6)、DHT干预组(D,n=6)、运动+DHT干预组(ED,n=6)和安静对照组(S,n=6),运动方式为20 m/min的速度、10%的坡度、60 min/d、持续10 d的跑台运动,DHT干预采用DHT缓释剂皮下埋植。最后一次运动后12 h取肝组织测IGF-1蛋白含量、AMPKαSer172和ACCβSer79磷酸化水平。结果:与S组相比,E组、D组及ED组肝组织IGF-1蛋白含量均显著升高(分别P<0.01,0.05,0.01),AMPKαSer172和ACCβSer79磷酸化显著下降(AMPKα:分别P<0.01,0.05,0.05;ACCβ:分别P<0.01,0.05,0.01);和E组相比,ED组IGF-1含量略有升高,AMPKαSer172和ACCβSer79磷酸化无显著变化。结论:耐力运动和/或DHT以IGF-1依赖性方式抑制大鼠肝组织AMPK/ACCβ信号;耐力运动与DHT共同干预对肝组织IGF-1和AMPK/ACCβ信号的影响无叠加效应。

      Abstract:Objective: The study examined the effects and effect mechanism of endurance exercise and/or DHT on AMPK and ACCβ of rat liver. Methods: 24 male SD rats were randomly divided into four groups: sedentary control (S, n=6), sedentary plus DHT (D, n=6), exercise (E, n=6), exercise plus DHT (ED, n=6). After all groups had been accustomed for one week, the group E & ED performed closely monitored training in the treadmill (grade of 10, at the speed of 20 m/min, duration of 1 hour per day)for 10 days. DHT’s slow releasing agent was used to D and ED group by subcutaneous administration. After 12 hours of the last exercise, liver tissue was isolated and IGF-1 protein content, the phosphorylation lever of AMPKα Ser172 and ACCβ Ser79 were determined. Results: Compared with the S group, exercise evoked a significant increase in IGF-1 level(P<0.01, 0.05, 0.01, respectively), and the phosphorylation of AMPKα Ser172 and ACCβ Ser79 significantly reduced in E, D and ED groups(AMPKα: P<0.01, 0.05, 0.05; ACCβ: P<0.01, 0.05, 0.01, respectively). Compared with the E group, IGF-1 protein content had slight elevation, but the phosphorylation lever of AMPKα Ser172 and ACCβ Ser79 had no significant change in ED group. Conclusion: endurance exercise and/or DHT inhibited AMPK/ACCβ signal in rats liver with IGF-1 dependence way; endurance exercise plus DHT had no synergistic effect on IGF-1 or AMPK/ACCβ signals.
