      摘要:目的:为了探讨力竭运动引起骨骼肌损伤过程中Caveolin-3、Myogenin和MyoD的作用,以雄性Wister大鼠为研究对象,采用一次性下坡跑运动,坡度为-16°,跑速16m/s,分别在力竭后即刻、6、12、24、48h和4、7d时取材。腓肠肌进行HE染色,测定血清肌酸激酶活性,RT-PCR法测定Caveolin-3、Myogenin、MyoDmRNA的表达。结果:骨骼肌形态学变化在损伤后24h、48h最严重;血清肌酸激酶在6、12、24、48h时均显著高于对照组,其中6h表达最高;Caveolin-3、MyoD和MyogeninmRNA均表现在先升高,后下降,其中MyoDmRNA在运动后12h达到峰值,Caveolin-3和MyogeninmRNA在24h达到峰值。结论:Caveolin-3、Myogenin、MyoD均参与到骨骼肌损伤及修复过程中,变化特点可能与三者在肌发生过程及肌损伤修复过程所起作用不同有关。

      Abstract: Purpose: to discuss the effect of Caveolin-3,Myogenin and MyoD during the rehabilitation of skeletal injury. Male Wisters were arranged to decline in the treadmill (-16° slope, 16m/s speed). Blood and gastrocnemius was obtained immediately, 6, 12, 24, 48h and 4 and 7days after one bout exhausting exercise. Serum CK was measured, gastrocnemius were HE dyed, Caveolin-3, Myogenin, MyoD mRNA were determined by RT-PCR. Results:The injury of gastrocnemius was the most seriously at 24h and 48h; Serum CK was the highest at 6h; Caveolin-3, Myogenin, MyoD mRNA rose firstly and decreased lately, but MyoD mRNA reached the peak at 12h. However, Caveolin-3 and Myogenin mRNA reached the peak at 24h simultaneously. Conclusion: Caveolin-3, MyoD and Myogenin took part in the process of the rehabilitation of skeletal muscle injury, their characteristics were perhaps connected with their effects during the myogenesis and the rehabilitation of skeletal muscle injury.
