      目的:观察8周有氧运动对大鼠心肌组织内Rho GTPases蛋白表达和mRNA转录的影响。 方法:符合实验条件的20只雄性Wistar大鼠随机分为安静对照组和有氧运动组,运动组的运 动方式采用跑台运动。以左室心肌肥厚指数作为判断心室重构形态学的指标;采用Western Blot法检测大鼠心肌组织RhoA、Rac1和Cdc42蛋白表达的变化;采用Real Time PCR观察8周 有氧运动对大鼠心肌组织RhoA、Rac1和Cdc42 mRNA转录的影响。结果:8周有氧运动下调大 鼠心肌组织内RhoA、Rac1和Cdc42的蛋白表达和mRNA转录,但是并未引起心室重构。结论: 运动对心肌内Rho GTPases的调控可能是心脏对运动锻炼产生良好适应的分子机制之一。
英文摘要:
      Objective: To investigate the effect of 8 weeks aerobic exercise on the left ventricular hypertrophy and the Rho GTPases in rat myocardium. Method: 20 Wist ar rats were randomly assigned to either a sedentary or an aerobic exercise group. The treadmill exercise is adopted by the aerobic exercise group. The level of ventricular remodeling were measured using the left ventricular hypertrophy i ndex (LVHI). The protein levels of RhoA, Rac1 and Cdc42 were measured using the Western Blot method. Furthermore, the mRNA levels of RhoA, Rac1 and Cdc42 were measured with Real Time PCR method. Result: 8 weeks exercise decreases the pr otein levels and mRNA levels of RhoA, Rac1 and Cdc42 but does not induce the ven tricular remodeling. Conclusion: Rho GTPases protein expressions and gene tr anscriptions in myocardium are mediated by exercise, which may be an important m olecular mechanism of adaptations of myocardium structure and function to physic al activity.
